Modern study releases a method in which amino acid is detected through a kit. This kit is used to detect a special amino acid which is as hydroxyproline. It is detected for the purpose of better food quality for which it is used as in food industry. Different color reactions are also used to detect it as ninhydrin. But it is more specifically used to detect peptides and proteins. The detected amino acid such as hydroxyproline is used as marker to detect specific diseases. 3-hydroxyproline is used to detect cancer like lung cancer or breast cancer etc. It is the form of hydroxyproline. 4-hydroxyproline is another form of it which is used to detect bone diseases. It is used to detect tumors and many other diseases etc. as a marker. The method of identifying of 3-hydroxyproline by a graph form of ninhydrin is not correct but some other amino acids can be detected by this method. It can be detected through a process which is known as HPLC. But it is not a pure method of its synthesis because of many columns. It is also very much time consuming method. But there is a need to separate hydroxyproline rapidly and easily nowadays. The method which is used to detect it easily is “detection through kit “. This kit specifically detect hydroxyproline-3 and 4. And it is identify by a process of absorbance and light spectrum in the form of curves. This process used chemicals in a solid phase. It also detects reagents of nitrobenzene. It also detect amino acid by the use of buffering agent.
Futher study produce another method of finding amino acid as firstly detect amino acid from nitrobenzene from solid phase. Then allow it to react with hydroxyproline and at last find it absorbance curve form due to this reaction. This reaction is time specific which tells that hydroxyproline is present or not. If it is present then nitrobenzene react with it. Solid phase contain buffering agent and an acid solution. The reaction which occurs between them provides us information about its structure. Other 20 amino acids can also be detected through this method. Detection of amino acid can also be detected by using different reagents and detection kit. Hydroproline is detected by a hydroxyl group. The 3, 4, 5-hydroxyproline found by this method. It is also used as a marker for disease detection. This method found L-form of it but further study also reveals its D-form. This found in solution and it contain water, glycerin etc. If the concentration of solution is higher than solvent is move form solution. While if it is low then released in the same way. While performing fluorescence spectrum a specific wavelength is adjusted. The same process is for absorption spectrum. When the two form of hydroxyproline is present in a solution then both can be detected because it cannot distinguish between the two used spectrums. It is found in urine sample. It is detected by using different reagents like; HPLC, L-proline.
Other amino acids can also be detected by different process like; spectrum and chromatography etc. The amino acid cannot move in a solid phase. In it different kinds of apparatus are used as test tubes, watch glass, beakers and other things etc. Also different kind of media is used in it. In different conditions it is collected as detection agent like; NBD-Cl. Hydroxyproline react with NBD-Cl and fined its reaction which occurs above. In order to maintain the solvent in which hydroxyproline is found which pH is maintain at 4. 0 – above 8. 5. It is maintained by a process of buffering agent which may be solid or liquid. The reaction time is about 2 mint. While it is also incubated at room temperature or at 20 to 50 C. In order to detect acids some inorganic or organic chemicals are used. This reaction occurrence ratio is 1:1. It is not depend on neutralization or any other type of reaction. The reaction product has 500 wave length. Its absorbance pick is about 500nm and in it no limit is applied. The absorbance spectrum have wavelength is about 350-660nm. While other have 250-550nm. The curve represents the presence of hydroxyproline and it is found through urine. Examples include formation of sample solution in which different amino acid are found. Drawing of reaction in the form of spectrum. By using all these methods hydroxyproline is detected without any manipulation. It is used in industries as examine the specimen at specific time. It is detected through reagents in Claim 1. While in Claim 2 it is detected through spectrum. It is detected through reaction in Claim 3. In Claim 4 it is detected through kit. Its L-form is used in the formation of medicines. It is prepared in cultural medium. It is synthesis from protein.
This method is not used in industrial preparation because it found in four isomer forms. It is prepare and get from animal tissues. Different steps are used to extract it. It is prepared in industry by the cultural media method. It is found to fulfil the requirement of the organisms. It is present in our body so if we take it as it is then it causes over dose of it so the better form of it is vitamin C. It is taken as a vitamin C. It is given to us because of its requirement in our muscles, ligaments, tendons etc. It is also essential for fish muscles etc. Collagen is a hydroxyproline and is not soluble in alkaline medium. It also exist in nature and found in feces. It is also a source of protein. It is soluble in acids. Its deficiency causes diseases like; compartment syndrome, polymyositis, myopathy etc. It also effect on ligaments, cartilage, cardiovascular diseases, venous insufficiency and respiratory diseases etc. It is also present in functional food, dairy products and health drinks etc. It require vitamin C as a cofactor. Collagen play role in its stability of triple helix. Trans-4-hydroxy-L-proline is the form of 4-hydroxyproline.
One of its form is used as in cosmetic compounds. These compounds provide advantages to the skin. It is good for dry skin. It protect our skin from radiations etc. N-acetyl-hydroxyl-l-proline is prepared from gelatin. It is use for the treatment of connective tissue disorder. It is obtained from hydroxyl-L-proline. Its separation method from others four forms is expensive. It is the normal component of many protein but gelation is used for many industrial preparations. It can also be prepared chemically but it has no importance. It is also prepare by a method in which arginine is removed from it but that method is very much expensive.
The main performance of this research is that arginine is not remove from it and it is less expensive and after that process good yield is obtained. More amount of yield is obtained as compared to above. Collagen contain polypeptides in it. It is composed of 300 repeats of tripeptide sequence. The single molecule of collagen is in the form of triple helices. The stability between them is not due to hydrogen bonding because it is not present in it. It is due to Water Bridge. It is used in the treatment of burns as tissue engineering. Collagen is also has variety of medical purposes. Collagen may be naturally occurring or may be synthesis. Collagen improves the strength. It also uses as biomedical devices. It also use to prepare semi collagen. Hydroxyproline differ from proline due to the –OH group. It has no use in therapeutic. It is present in blood naturally. It is also present in vegetable oil based products, frozen products. It is also used as a meal replacer. Hydroxyproline is used as an administrator of routes as in therapeutic composition. Its administration may consist of single route or multiple routes. It is also use as dosage of about 0. 01 to 1000mg. hydroxyproline is important for humans, children, pregnant women, an adult, an intact, a man, animals etc. It is also essential for an individual that is healthy, ill, weightlifter, bodybuilder etc. Examples include effect on muscles tissues, on connective tissues, on lungs function, on edema on joints ligaments and tendons etc.
It is used to improve the quality of meat etc. and also improve the texture of firmness etc. It is also use as a feed composition. It may be raw material or a meat or any other diet. It can also be given as a dietary supply as vitamins A, B, C etc. Its highly deficiency causes gapping. Its N and C terminus form cross link formation which is also known as gyration. It is needed by the animals 3 to 5 times daily in oral form. Humans need it 3 to 8 times daily. The major role of L-form of it is in inflammations. It is also a sweetening agent.
